according to TV.com's episode guide the 100 episode is called Stewie kills Louis, the 101st episode is called Louis kills Stewie.
I think it's Louis as she is not mentioned in any later episode summaries at this time, but that's just my thoughts.

Sounds like an urban myth in the making to me. You "heard" that someone would die in episode 100, but you don't seem to have any concrete details or any explanation for why you seem to believe the rumor.
Given that we're discussing an irreverent cartoon, I wouldn't take the rumor very seriously. Even if there were plans to kill off a character for publicity, which I wouldn't rule out entirely, I find it hard to believe that the death would be lasting or meaningful. Personally, if the writers feel that they need to kill off someone, and I don't have much faith in that theory, I rather hope it would be Kenny from South Park, since he's used to dying.
It will be interesting to see if the rumor will continue to spread or not; either way, the rumor should be good for the show, since any publicity is good publicity.
Episode 101-102 is a 2 part episode called "stewie kills louis" apart from that there is not much of a description.
I'm pretty sure its Lois, and Stewie kills her because she left him behind while she went on a cruise with peter. There's an episode latere title "Anchorwomen: The legacy of Lois" so i think she dies.
Lois is killed by Stewie. Stewie gets pissed that Lois leaves him to go on a cruise so Brian challenges Stewie to actually do it. I hope Lois isnt gone from the show.
