To an extent he does. He doesn't get tired easy and it is difficult to hurt him, but yes, he does have a healing factor.
Superman is capable of holding his breath for a long time and is capable of holding massive amounts of oxygen in his lungs.
I believe that Superman does have a healing factor which is related to the absorption of yellow-sun radiation. The more yellow-sun radiation he absorbs, the faster he heals.
Superman can't breathe in space, but he can go for very long periods of time without breathing. But if he wants to take a long journey in space, or be able to communicate with others, he needs to wear a space suit with an oxygen supply.
