You definitely have to watch Fullmetal Alchemist. It has all the qualities of an excellent anime. It's packed full of humour, adventure, fighting, drama, and it deals with very mature issues like death and resurrection. It is definitely an anime that should be at the top of your list to watch! =)
Another good anime that fits into those categories is Scrapped Princess. It's somewhat like Fullmetal alchemist in that it deals with mature/serious issues, and it also has very good fight scenes, and it's just a great watch overall. I recommend you watch this anime too. =)
