ok heres the real answer ....tsunade is the first hokage's grand daughter and tsunade is the 5th hokage tsunade's bot fren don(dead) is shizune's uncle also they are not related but they COULD of been in-laws if tsunade and don got married also shizune is tsunades assistant and later on in naruto sakura also becomes tsunades assistant

who are you talking about? do you mean tsunade, and if Shizune is her assistant girl, then they know each other because Tsunade's old boyfriend was her uncle. I'm not sure if that's who you're talking about, but I hope so.
No. Tsuade was the first hokages granddaghter and the second hokage was shizune's uncle. So they aren't related. Though they sort of are more like friends even though shizune is tsuade's assistant.
