How about a woman who could shape-shift into other females, or at least be a master of disguise. Then they could surprise him by appearing as MJ Watson, Gwen Stacy, Aunt May, etc. Kind of like Mysterio in the old days, but not hallucinations.
Or, one thing I have never seen done is a villain who for whatever reason does not stick to his web.
Try to create a foe that has traits not seen before that SpiderMan would have trouble dealing with.
maybe someone who can control technology with his mind.

why not have an alien who could morph into anyone and have their powers and abilities but it would not work if the person powers were artificial that way she could beat him at his own game or beat him with someone else's game and he would have no idea whose powers she would be using at any time and you could name her
