Girl I love that show!!! OK here are some ideas:
1. Have your little girl be "Tyra" and have her set up different tasks on postcards. ie if your girls name is "Tracy", make up little cards that say "Tracy Mail". Each postcard can have things like " you may be model material, but can you show it on paper?" then have the girls draw your child in her best model pose. Best picture wins a prize.
2. Have a red carpet going from your front door to the main room where the party is.
3. Have little stations set up around the party area: makeup, different accessories, and disposable cameras. Have them get an outfit together with makeup in a time frame (5min or so), then have them do a walk on the red carpet while you and other guests snap away with the cameras! Best outfit wins a prize!! You get the idea, now go from here!
Forget the cake and ice cream, obviously. And instead of "pin the tail on the donkey", you could have the kids play "throw up the iceberg lettuce you had for lunch." It's pretty warped to throw a party for a 12 year old that's themed around a show that judges contestants' weights and appearances. At 12, kids are starting to go through lots of changes, and most are feeling pretty unsure of their bodies as is. Why call more attention to appearances?!
Make yourself a "runway" by setting chairs up facing each other about 4 feet apart. Have each "contestant" walk the runway:
-in a pair of VERY high heels
-with a book on her head
-with a blindfold on
Have the girls divide into teams (you may need to draw names to avoid squabbles) and give them assigments, such as:
-they can choose one girl to do makeup on
-choose another to do a hairstyle on
- choose another to model an outfit made out of common items (toilet paper, scotch tape, newspaper)
You can vote and have winners, or you can just do it for fun.
Lastly, I would have each girl who is willing come up with something in the way of a "talent" like a beauty contest does. You could use a karioke machine for this.

PS If you the whole thing on video tape, you will give them LOTS of entertainment.
