How old a movie? I hope you're not thinking of Tiny Tim who played the ukulele like on the Tonight Show. He did appear in one movie called Blood Harvest (1987).
But the first actor from the old days that I thought of was Maurice Chevalier. He was well known for playing the ukulele back in the 40's and 50's.
